Formulation of natural hydrogel from bromelain enzym and alginate-chitosan and the effectiveness test by in vivo in healing burns

Abstract

The causes of burns vary, ranging from low levels of work safety, high levels of traffic accidents, or carelessness in household activities, and cases like this are still quite common in developing countries. The impact felt by burn victims can be mild to very severe depending on the degree of burns suffered. Severe burns require additional external growth factors and matrices to induce a natural healing response. The bromelain enzyme that can be found in all pineapple tissues and consists of anti-inflammatory properties can be an alternative in accelerating burn healing. This study aims to test the effectiveness of a combination of alginate and chitosan hydrogel preparations containing the active enzyme bromelain in accelerating burn wound healing. This study began by optimizing alginate-chitosan as the gel base that was formulated in three variation concentrations. Physical evaluation of the gel was applied to all formulas which include organoleptic test, viscosity test, pH test, spreadability test, adhesion test, homogeneity test, and irritation test. The in vivo test was conducted to assess second-degree burns in male white mice (Mus musculus) divided into five groups, namely the treatment group of F1 (0.1%), F2 (0.5%), and F3 (1%), the positive control group of Bioplacenton® gel, and the negative group control of alginate-chitosan gel base without active substance. The physical evaluation test results showed that all formulas met all requirements while the effectiveness test results showed that the F3 1% formula was the fastest formula in accelerating burn healing with p < 0.05. © 2025 by SPC (Sami Publishing Company).
